Apoptosis in Denervated Skeletal Muscle International Journal of Morphology
Davatz,Giovanna Castilho; Andreo,Jesus Carlos; Rodrigues,Antonio de Castro; Rosa Júnior,Geraldo Marco; Moraes,Luiz Henrique Rapucci.
The apoptosis phenomenon happens in normal tissues and in many pathological conditions as described by several researchers. Most studied in the 70s, this phenomenon remained almost forgotten in the 90s, when, with the advent of more advanced techniques, returned to laboratories. Thus, electronic microscopy, Comet Assays and histochemistry techniques, among others, were utilized in an effort to elucidate in a thorough way, the mechanisms that lead the cell to "suicide". No one can deny the importance of the knowledge of such phenomenon, so as to control its induction or blocking in the treatment of pathologies such as cancer. In this work, the apoptosis study was aimed at denervated muscles in different periods. Furthermore, two types of skeletal striated...

Choline acetyltransferase detection in normal and denervated electrocyte from Electrophorus electricus (L.) using a Confocal Scanning Optical Microscopy Analysis Anais da ABC (AABC)
NUNES-TAVARES,NILSON; CUNHA-E-SILVA,NARCISA LEAL; HASSÓN-VOLOCH,AÍDA.
Acetylcholine is the neurotransmitter responsible for the transmission of impulses from cholinergic neurons to cells of innervated tissues. Its biosynthesis is catalyzed by the enzyme Choline acetyltransferase that is considered to be a phenotypically specific marker for cholinergic system. It is well known that the regulation of Choline acetyltransferase activity under physiological and pathological conditions is important for development and neuronal activities of cholinergic functions. We observed the distribution of Choline acetyltransferase in sections from the normal and denervated main electric organ sections of Electrophorus electricus (L.) by immunofluorescence using a anti-Choline acetyltransferase antibody. The animals were submitted to a...

Ciliary Neurotrophic Factor (CNTF) affects the excitable and contractile properties of innervated skeletal muscles Biol. Res.
RAMIREZ,BEATRIZ U; RETAMAL,LEIRA; VERGARA,CECILIA.
The well-established trophic role of CNTF upon neurons led to performing clinical trials in patients of neurodegenerative diseases. However, trials were suspended due to side effects such as severe weight loss, hyperalgesia, coughing, muscle cramps and pain. So far it is not known how CNTF triggers the problems related to skeletal muscle cramps and pain. CNTF has also been described as a myotrophic factor for denervated skeletal muscles, but the possibility that it affects innervated muscles has also been considered. Since a myotrophic factor could be a valuable tool for treatment of several muscle diseases, we studied the effects of low doses of CNTF delivered systemically by an osmotic pump, over the electrical and mechanical properties of innervated and...

Correlation between Innervation of Skeletal Muscles and Myoblast Stem Cells International Journal of Morphology
Wu,Jiang; Sun,Xiaojuan; Zhu,Min; Zhu,Chongtao; Chen,Bin; Xu,Yongqing; Ouyang,Jun; Liao,Hua; Qiu,Xiaozhong.
In order to explore the change rule of myoblast stem cells (satellite cells, SCs) in the denervated and re-innervated muscle and to investigate the cellular mechanism of the morphological and functional changes of the muscle, denervated muscle atrophy and nerve regeneration models were established in one-month-old rats. Postoperative indexes such as muscle wet weight, cell section areas, content of collagen fibers and DNA, electrophysiology, numbers of SCs in the triceps muscle of calf were dynamically tested. After denervation, the muscle wet weight and cell area reduced rapidly, and the collagen fiber content increased slowly. The number of SCs increased at first, and then declined suddenly two months later. From 4 to 5 weeks after re-neuralization,...

Effect of salbutamol on innervated and denervated rat soleus muscle BJMBR
Šoic-Vranic,T.; Bobinac,D.; Bajek,S.; Jerkovic,R.; Malnar-Dragojevic,D.; Nikolic,M..
The objective of the present investigation was to perform a 14-day time-course study of treatment with salbutamol, a ß2 adrenoceptor agonist, on rat soleus muscle in order to assess fiber type selectivity in the hypertrophic response and fiber type composition. Male Wistar rats were divided into four groups: control (N = 10), treated with salbutamol (N = 30), denervated (N = 30), and treated with salbutamol after denervation (N = 30). Salbutamol was injected intraperitoneally in the rats of the 2nd and 4th groups at a concentration of 0.3 mg/kg twice a day for 2 weeks. The muscles were denervated using the crush method with pean. The animals were sacrificed 3, 6, 9, 12, and 14 days after treatment. Frozen cross-sections of soleus muscle were stained for...

Effects of long-term fluoxetine treatment on adrenergic plasticity in rat vas deferens BJPS
Souza,Cláudio Fontes; Garcez-do-Carmo,Lúcia; Conceição,Isaltino Marcelo; Freitas,Thalma Ariani; Frussa-Filho,Roberto.
Chronic antidepressant administration increases neurotrophin levels in the central and peripheral nervous system, leading to an increase of neuronal sprouting, reestablishment of neural networks and neurotransmitter levels. Injured peripheral nerves regenerate at very slow rates. However, the recovery of the hypogastric nerve in rodents after injury is significantly improved with neurotrophin administration. Accordingly, our goal was to determine whether treatment with the antidepressant fluoxetine affects catecholamine levels and neuronal function, after surgical denervation of the rat vas deferens. Noradrenaline levels in the denervated vas deferens were higher in fluoxetine-treated animals than in the vehicle-treated group, as measured by high...

Glucose metabolism in rats submitted to skeletal muscle denervation BABT
Nunes,Wilton Marlindo Santana; Mello,Maria Alice Rostom de.
This study analyzed the local and systemic effects of immobilization by denervation of the skeletal muscle on glucose metabolism. The rats were submitted to section of the right paw sciatic nerve. A reduction was observed in glucose uptake by the isolated soleus muscle of the denervated paw after 3 and 7 days, but not after 28 days in relation to the control animals. There was no difference after 3 and 7 days in glucose uptake by the soleus muscle of the opposite intact paw in relation to the control. There was increased glucose uptake in the same paw 28 days after denervation. The rate of glucose removal in response to exogenous insulin after 28 days of denervation was significantly higher than in control animals and those observed after 3 and 7 days of...

Implantation of muscle satellite cells overexpressing myogenin improves denervated muscle atrophy in rats BJMBR
Shen,H.; Lv,Y.; Shen,X.Q.; Xu,J.H.; Lu,H.; Fu,L.C.; Duan,T..
This study evaluated the effect of muscle satellite cells (MSCs) overexpressing myogenin (MyoG) on denervated muscle atrophy. Rat MSCs were isolated and transfected with the MyoG-EGFP plasmid vector GV143. MyoG-transfected MSCs (MTMs) were transplanted into rat gastrocnemius muscles at 1 week after surgical denervation. Controls included injections of untransfected MSCs or the vehicle only. Muscles were harvested and analyzed at 2, 4, and 24 weeks post-transplantation. Immunofluorescence confirmed MyoG overexpression in MTMs. The muscle wet weight ratio was significantly reduced at 2 weeks after MTM injection (67.17±6.79) compared with muscles injected with MSCs (58.83±5.31) or the vehicle (53.00±7.67; t=2.37, P=0.04 and t=3.39, P=0.007, respectively). The...

Morphologic Alterations Resulting from Denervation of the Diaphragm in Rats International Journal of Morphology
Torrejais,M. M; Soares,J. C; Matheus,S. M. M; Francia-Farje,L. A. D; Mello,J. M; Vicente,E. J. D.
The morphological alterations that occur in the muscle fibers of denervated rat diaphragms were studied. Fifteen adult male albino rats (Rattus norvegicus) with a mean weight of 200 g and about 60 days of age were used. Chronically denervated diaphragms were obtained and the animals were sacrificed after 4, 8 and 12 weeks of denervation. The left antimere of the diaphragm was denervated by sectioning of the phrenic nerve and the right antimere served as control. Each antimere was divided into two fragments, which were used for histological (H.E.) and histoenzymological (NADH-TR and myofibrillar ATPase). After 4 weeks, denervated muscle fibers showed important light microscopic alterations: atrophy with angular profiles in cross-sections, cytoplasm...

PPM1B and P-IKKβ expression levels correlated inversely with rat gastrocnemius atrophy after denervation BJMBR
Wei,Jian; Liang,Bing-Sheng.
Activated inhibitor of nuclear factor-κB kinase β (IKKβ) is necessary and sufficient for denervated skeletal muscle atrophy. Although several studies have shown that Mg2+/Mn2+-dependent protein phosphatase 1B (PPM1B) inactivated IKKβ, few studies have investigated the role of PPM1B in denervated skeletal muscle. In this study, we aim to explore the expression and significance of PPM1B and phosphorylated IKKβ (P-IKKβ) during atrophy of the denervated gastrocnemius. Thirty young adult female Wistar rats were subjected to right sciatic nerve transection and were sacrificed at 0 (control), 2, 7, 14, and 28 days after denervation surgery. The gastrocnemius was removed from both the denervated and the contralateral limb. The muscle wet weight ratio was...

Study of the myenteric and submucous plexuses after BAC treatment in the intestine of rats Biocell
Buttow,Nilza Cristina; Santin,Miriam; Conci Macedo,Luciana; Neres Teixeira,Aline Cristina; Novakowski,Gisele Caroline; Bolonheis Armelin,Taíse Roberta; Assmann,Kathya.
A morphological and quantitative study in the ileal and colonic myenteric and submucous plexuses of rats after BAC denervation was performed. Four groups were employed: SI - ileum control; CBI - denervated ileum; SC - colon control; and CBC - denervated colon. We used the Myosin-V immunohistochemistry technique to study the myenteric and submucous plexuses. In the submucous plexus of the ileum and colon there was not a significant decrease in the number of neurons/mm2 and of ganglia/mm2. The denervation of the myenteric plexus in the group CBI was 44.7% and in the group CBC, 68.3%. In the myenteric plexus there was also a significant decrease in the number of ganglia/mm2 (13.8% in group CBI and 52.14% in group CBC) and in the number of neurons/ganglion...
